The author’s purpose in writing this article was not to show the â€Å"Nacirema† as an example of how extreme human behavior can become, but how an outside perspective can affect your perception of an alien culture. If one were to look at the â€Å"Nacirema’s† cultural behaviors regarding physical appearance and health without any insight or knowledge of the specific beliefs or values of that culture, they might seem bizarre and even incomprehensible. In The Great Gatsby, when Mr.Gatsby told Vick he wantedto return the past over again with his lover- Daisy, Nike Carroway warned him to give it up, because it was impossible. Unforturately, Mr.Gatsby was not believe it. So at the end, Mr.Gatsbys dream still had not came true because Daisy did not break up with Tom and go with him. By these facts, he had a clear mind and reliable in his observations, so it can make people believe Nike Carroways was a neutral and clear mind narrator of this novel, because this can be seen in Nikes personality. In the novel, Nike was also a neutral narrator too becasue he is the narrator who described everything clearly and accurately. He was trustful because he described everything without any personal point of view; By the way, throughout the whole story, he didnt defenses for any characters nor put any of his self-feeling in it. It was paid for by the African National Congress (ANC) who were strong supporters of Mandela and the African civil rights movement. This means that it may have been biased in favour of the civil rights movement. The ANC wanted to show how badly mistreated the blacks in Africa were in order to gain support from many countries around the world and defeat apartheid. It was printed in 1988 when there was already quite a lot of support for the movement following the Free Mandela song and concert in 1984. This means that the ANC were probably also trying to reinforce the peoples view of apartheid and Mandelas imprisonment. The source is quite limited and although a primary source does not give any information on the suffering of the ordinary people at the time. The source also has a wide range of data ranging from the population to the average salary of both blacks and whites. It does however not show how the actual living conditions were in South Africa for the blacks. The advert also strikes at the government of South Africa and claims that it is outlawing democracy. They believed that Mandela was a figurehead of democracy and freedom. They wanted to show him and the movement in as good a light as possible. Source D is a short extract from a speech made by the South African Minister of Information. This title is rather like Josef Goebbels in Germany under Hitlers reign. He masterminded all of the propaganda, as does the South African Minister of Information. This makes the source extremely biased towards the whites. The writer of this speech would have wanted to show Mandela and the rights movement in a bad light to destroy its support. The government also wanted to deny the civil rights movement and its pursuit as a valid political choice. They took advantage of the acts of terrorism by Mandela and other blacks. This showed that the blacks were not at an equal level and that they were also not going about their pursuit the right way. If the government could portray Mandela as a terrorist they could remove a lot of the sympathy he had gained from all around the world. This is done by stating that Mandela is not in prison because of his politics but that he is guilty of a serious crime. This was quite a successful argument against Mandelas release and contrasted greatly to the information received from the ANC. Both of the groups had different reasons for supplying these reports and they contradict each other so that it is difficult to know who is telling the truth. 4. Source E is from a British newspaper, and is a cartoon drawn by a British cartoonist criticising Apartheid. It was drawn in 1964, which was very early in the African civil rights movement.

To initiate breeding condition of female quails, the researchers placed them inside a stainless-steel cage for a maximum of 30 days. On a daily basis, each male quail was paired with a couple of randomly selected female quails until the male quail would mount the two females for two weeks. Sounds from conversations hum in the air around them, the clinking and bustling of a busy cafà © a background to this conversation. Despite the multidimensional setting—the music playing in the speakers above, the couple to their left discussing baby names, the loud ring of the nearby telephone and the even louder server who answered it—these two talk as if they were alone at home, as if none of the loudness existed. They are celebrating. Crumbled remains of a chocolate fudge cake occupy the table space they share, and two large mugs of coffee sit pushed to the side, ready to be taken away.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   This is where a server makes it big or loses out completely on a tip. Time is a factor in everything, and as a server you have to be willing to divide your time evenly among your tables so that you don’t fall in the weeds later. If you see one of your tables finished with their plates and cups, it’s your job to pick it up, and fast. It’s an opportunity to offer them anything else, and one more chance to make an impression, to make them like you. They’ve finished eating, and more importantly, she’s just eaten chocolate cake.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   So I bustle over and say, â€Å"Hey folks, can I get this out of your way?† Smiling, of course. You have to smile.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   She looks up at me, blinking, cross, brow furrowed. He moves forward, sitting up aggressively, brow furrowing, â€Å"What, man?†   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   â€Å"Can I get this out of your way?† I ask, cautiously, not sure what just happened here. I even lean in a little, my hands crossed behind my back so I look more servant-ly, less threatening.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   The mood cracks. She laughs. He smiles goofily in relief. He shakes his head. She puts her hand over her eyes. â€Å"We thought you said something else,† they both seem to say at the same time.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   When they leave, I find my tip—a measly $3—under the saltshaker. Apparently, noise is a factor, too. The external part of it, all those people’s voices charging the air around us, music swirling in between conversation, me asking to clear the plates; they thought I was hitting on her. Maybe I was smiling too much.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   But he must have that internal part of it, the voices in his head that tell him she’s cheating on him with every guy that sees her. Why is that, I wonder? They were having such a nice conversation, and then I got there, and everything changed. What were they talking about? Being a server is about the closest thing you can be to a fly on the wall. They almost ignore you completely, until you buzz around them and ask if it’s okay to pick up their plates. Next time, I shouldn’t say anything, I should just pick it up.

Human Nature in Bartholomae and Petrosky's Our Time, Theft, and Music of the Swamp Why should college students read the stories that are assigned in English courses? Other than to satisfy the professor, what is the purpose of reading these difficult writings of people we don't know or care about? Many of these students find themselves asking, "What is this writer talking about?" Confused, some quickly give up trying to understand the story and make reading something just to get through, diminishing both their understanding and their grade. Knowing what these writers are trying to explain makes their stories much easier to read. Throughout history, we humans have tried to understand why we do the things we do. To aid in our understanding, many storytellers throughout literary history have written fictional and non-fictional stories about human nature to help others, as well as themselves understand. Human nature is what the writers of Our Time, Theft, and Music of the Swamp, three excerpts from the anthology Ways of Reading edited by David Bartholomae and Ant hony Petrosky, often read in English courses, are trying to explore. My personal story, Chinese Food Can Save Your Life, written for my English composition course is also an example of this exploration. The human nature in these stories is to blame other people, places, or situations for failures and general unhappiness. Most readers can probably relate to this since at one point or another, they have thought that, if they just had some extra money, a better job, a different lover, a new home, or a better childhood, they could be happier. To assign the blame to other people and things is easier than to point the finger at ourselves. Although a few things individuals are not responsible for do exist, such as ethnicity and hereditary characteristics, most of the things good or bad that happen to us are a result of choices we have made. In these stories, this human compulsion to obsess for what we (supposedly) don't have destroys any possibility of obtaining the particular possession. In Theft a chapter from Joyce Carol Oates' novel Marya: A Life, the main character Marya blames dependency for her unhappiness. Early in Marya's life she decided that dependence on other people and involvement in relationships resulted in her limited freedom.

Negligence Paper Ann Fairvalley University of Phoenix HCS/ 478 Negligence Paper Imagine waking up in the recovery room from being sedated for a procedure in which one of your limbs has been amputated. While in recovery you are in and out of consciousness. Finally after being in recovery for 2 hours you are taken to a step down unit to recover and receive teaching and therapy. After getting settled into bed you gets the guts to throw back you sheets and take a look where there was once a left leg.To your horror your left leg is there and your right leg is not! â€Å"The Joint Commission considers wrong site surgery to be a sentinel events which is defined as â€Å"an unexpected occurrence involving death or serious physical or psychological injury, or the risk thereof. Serious injury specifically includes loss of limb or function† (Dillon. 2008. ). † This was an all too familiar of a situation for one Mr. Joseph Benson a 62-year-old diabetic with circulation problems tha t required a leg amputation.In this paper I plan to explore the legal implications in regards to the differences between negligence, gross negligence and malpractice. I will also explore some rational as to why union problems and/or nursing shortages could have been the cause of this disaster. Lastly I will describe the importance of documentation in regards to potential negligence as if I was the nurse involved with Mr. Benson’s care. I will briefly describe my ethical principles, which would guide this practice and how I would document the case to satisfy ethical and legal requirements. Negligence as defined by Black’s Law Dictionary (1979) and the Joint Commission on Accreditation of Healthcare Organizations (JCAHO, 2002) is the â€Å"failure to use such care as a reasonably prudent and careful person would use under similar circumstances. † It is the predominant theory of liability in medical malpractice litigation (King, 1986) (Weld. Garmon. Bibb. 2009. ). † During my experience as a surgical nurse working in Obstetrics we used checklists and timeouts to make sure that we followed hospital protocols during our procedures.The checklists were in place to make sure procedures and paperwork was completed in an orderly and prudent manner. We could not continue onto the next step in the checklists until it was crosschecked by another licensed nurse or physician. This was put in place to ensure the patient’s well being and safety was addressed at each step in the checklist. Each one of the JACHO accredited hospitals has a similar checklist in place to ensure the patient’s wellbeing and safety. By having timeouts and checklists this could possibly prevent negligence. Gross negligence means, â€Å"reckless indifference to or a deliberate disregard of the whole body of stockholders† or actions which are â€Å"without the bounds of Reason (Sharfman. 2006. ). † One example of gross negligence would be giving a pa tient a higher dose of a narcotic instead of wasting the unused portion in order to keep the patient quiet and sleeping during a hectic shift. This nurse would have made a conscious decision to heavily sedate her patient against doctor’s orders, which would constitute gross negligence on the part of this nurse in this situation. In the case of Mr.Benson I think the physician could have been found guilty of gross negligence. If the doctor in this case would have followed timeouts and checklists the wrong procedure could have been avoided. â€Å"Barron’s Law Dictionary (1991) defines malpractice as a professional’s improper or immoral conduct in the performance of duties, either intentionally, through carelessness, or through ignorance (Weld. Garmon. Bibb. 2009. ). † The physician in this case could also be found guilty of malpractice. His neglect of professional duty comes in question as to the welfare of Mr.Benson. Did the physician show a lapse of judgme nt in his conduct during the time prior to the actual surgery by not following the procedures and protocols? It was not documented in the article as to the physician’s demeanor or conduct during the actual procedure. I do not have a written account of the scrub nurses or other Operating Room staff testimony as to what actually happened in this Operating Room. It would be speculation on my part to assume the worst of the physician without having all the details before me. I choose to agree with the patient. Mr. Benson was orrified at the outcome of his procedure. I would not matter to me whether the Neighborhood Hospital had nursing shortages or problems with the unions all I would be concerned with is how could this happen and what is the hospital going to do about this mishap. I would contact an attorney who specializes in Malpractice litigation and see what my options were as far as litigation against the hospital and also the physician. I would research about sentinel even ts and JACHO regulations which could further my case against the hospital and physicians who were involved.It is very important to document each and every step and to narrate in written form what happens while the patients are under your care. In the day-to-day world of nursing we have a saying â€Å"If it wasn’t documented it never happened. † I have never forgotten these words. It made a resounding impression on my professional nursing practice. Good, bad or ugly I document what actually happens during my shift and my care of my patients. At times, this includes having to document physicians demeanors, actions or lack of.I have had to document that I made several attempts to reach a physician by phone and left messages on his answering service and voicemails in which he did not return phone calls in a timely manner. It can be very frustrating as a professional nurse when this situation occurs as you are put in a place in which you are against a rock and a hard place. Nonetheless, you have to document to protect yourself and your patients. In conclusion I have defined negligence, gross negligence and malpractice. I have given personal stories of how these terms applied to what I have seen in my professional nursing career.I agreed with Mr. Benson and his position and did not agree with the rationale of the Neighborhood Hospital for the injustice, which happened to Mr. Benson. Lastly I described the importance of documentation in the role of a nursing professional. I would hope that the next time I need a surgery or a minor procedure that I can trust my hospital and the staff to perform as they should, this would include following hospital procedure and protocols as well as follow there professional standards correctly.References Dillon, K. Seeing Things from Someone Else’s Perspective

In your everyday life, you are placed in situations that allow you to encounter the value of carefully considering someone else’s perspective. The reason why it is so eulogized to see someone else’s view is because it could alter how things would eventually turn out to be for you and that person(s). Peace, equality, and justice are some of the values you come across when you approach with empathy. Comprehending where someone is coming from and feelings they convey correlates with the outcome of peace.In 1993, the ongoing conflict between Israel and Palestine had temporarily halted with a solution called the Oslo peace process, which main point consisted that Israel gradually give up possession of Palestinian territories to the Palestinians with the exchange of peace. This represents the value of perspective, because the Palestinians had finally reached the conclusion that coming at the Israelis with violence and weaponry would not allow the Palestinians to ever gain wha t they really want, and it caused the Palestinians to comprehend what the Israelis were feeling; harassed.They had learned that if peace were used as a substitution to attack, then the people of Israel would be sure to ably, which they transitorily did. Another value would be justice. When an attorney is defending their client, they tend to use the skill of switching to the defendant’s perspective when trying to prove to the jury that they are not guilty. For example, if someone was falsely accused of murder, the attorney could use empowering words and point things out like â€Å"Just think if you were in his/her place† or â€Å"How do you think you would’ve felt if you were him/her? to give the jury that connection that â€Å"Would you want to be proven guilty if you were to fall under a situation similar to this one? † Using this technique shows how perspective comes in handy as well as how justice is served. Equality is a reiterating value that is dis covered when considering someone else’s perspective. In Remember the Titans, a film about how an African American named Herman Boone who is newly appointed head high school football coach in the position of a successful white coach, Bill Yoast, during the time of segregation, there is a scene where Coach Yoasts’ daughter, Sheryl s over at the Boones’ house when an anonymous student from the school drives by and yells â€Å"Hey, Coach Boone! † in a tormenting manner and throws something at their window near to where Sheryl had been sitting, shattering it to pieces. Later that night Yoast tells Coach Boone that he is not in any position to put his daughter’s safety on the line just because people don’t like him coaching the team, to which Boone replies â€Å"Well what about my girls’ safety?You just got a taste of what I deal with everyday, Yoast† leaving Coach Yoast thoughtful and silent. This also shows perspective because Coac h Yoast had understood that the worry and the vexation, that had aroused from the danger of his daughter is a mental weight and distress that Boone has to deal with for his family all the time, all because of the color of his skin. By stepping into Boone’s shoes, Yoast had now truly known the importance of equity and fairness to Boone, and the rest of the African American society.These are just a few ethics you can obtain from point-of-view. Agreement between even two nations can come about when using perspective. Authority can be placed with shifting someone’s point-of-view through sympathy. And adequation will make its way through when you are physically put forth into the same situation that someone who isn’t treated fairly is. All of these are real and pure values that form when your mindset is reversed, and truly leave you thinking.

Stomp The Yard Review Stomp the Yard is by far one of the best captivating and entertaining movies I have ever watched. If there is a movie which I can vote for completely, then it is the movie stomp the yard. This movie is about an inner city krump dancer, DJ, who moves to Atlanta, Georgia for a fresh start. In the movie, DJ’s role is arrogant, over-confident, and determine to win the heart of April Palmer. The plot of the movie is so entertaining and touching that I could watch it everyday. This is my favorite movie because it focuses on responsibility and determination, it also shows how one person can not be above everyone else, it takes teamwork. Lastly, the movie shows how a man is to appreciate and treat the lady he loves. Stomp the yard focuses on competing fraternity steppers at fictional Truth University. DJ channels his anger at the system though crunk dancing, and his innovative moves make him a crowd favorite. But when a fight with a rival team leads to his brother Durons murder, DJ feels guilt and despair, which turns, no surprise, into more anger. Shipped off to Atlanta to live with his uncle and aunt, DJ finds new athletic, dance, and competition possibilities in stepping. At Truth, DJ is recruited by two fraternities who think his skills will help them win the national championship. He selects Theta Nu Theta because its leader, the one and only, Ernest Sylvester extols the virtues of brotherhood more than winning the title which of course, everyone focuses on winning. I give full credit to the director, producer and editors of this film. They really kept their brains working when they were coming up with all the ideas composed of the film. Such ideas as stepping, DJs love for April, Grants rivalry with DJ, April fathers grudge against DJs uncle for having stolen his girlfriend back in the day and the Pythons dream to beat The MU Gamma at the National championships. This film is well directed. For instance, when DJ moves to Atlanta and joins The Truth University, he discovers, stepping, the dancing technique which is traditionally done in African-American fraternities. It is here where teams demonstrate their complex moves and create rhythmic sounds by the use of their bodies. For example DJs talent sees him at the peak of hip-hop between rivalry of the two fraternities (Pythons and MU Gamma), the winner of which will be determined in front of a sold-out-arena at the annual championships, nevertheless, DJ has to overcome his own ego and learn the true meaning of brotherhood. The films nature has its audience fully involved. The editing of this movie was done with the highest perspective. Its rhythms are well edited, they are captivating and entertaining. In depth Stomp the Yard demonstrates responsibility and determination that makes this movie so real and interesting. The costumes and make-up directors did a marvelous job. The fact is that the costumes used in the movie were captivating. The costumes added relevance to the motion and main ideas in the film. For example, at the last stage of the movie, The MU Gamma fraternity presented a show that incorporated wolf sounds. This was further enhanced by the fact that the group had worn torn clothes. The use of different costume by the different fraternities brought out their sophistication. The director selected costumes that are widely used by todays generation of dancing artists. This film has a significant impact on its audiences. There are several aspects that have been tackled in the movie. The movie develops and improves on the dancing skill of those interested in dancing. It also urges the individuals (specifically the youth) to fight for what they want through dedication and commitment. For example, DJ has to overcome so many obstacles in order to accomplish the goals he has. The movie also shows the importance of standing by friends and family. This is shown when DJ is taken in by his uncle and also when April stands up to her father in order to defend DJ. April also comforts and encourages DJ when hes just about to quit and she manages to convince him to keep going. Â  Furthermore it portrays the consequences of hatred among individuals. For instance, the hatred that Aprils father has for DJs uncle is immense such that he does not want DJ to be with his daughter. The hatred between DJ and Grant also causes DJs suspension (when grant investigates abou t DJs past and finds that he had a criminal record). For those people that love dance movies, stomp the yard brings new ideas and moves to the screen. The movie unlike other many dance films enhances the importance of uniformity when performing as a crew. It is a truly captivating film that is fun and educative to watch. Heavy-handed and well- intentioned, Stomp the Yard proposes that step groups (and similar organizations) provide structure and inspiration for students in need of guidance and a sense of belonging. Its a decidedly masculine melodrama. Not only does DJ contend with paternal dis approvals (his uncle thinks stepping is a waste of time, and Aprils dad warns him that My daughter is not some shorty for you to mess with!), but he must also come to terms with his own competitive hostility and Durans death. At the same time, he has to come up with a killer step routine for the group. Luckily, DJ is inspired by Truths amazing array of sorority and fraternity alumni, who are enshrined in Heritage Hall, Â  the list includes Esther Rolle, Hines Ward, Michael Jordan, Rosa Parks, and Coretta Scott King, among others. Â  Stomp the Yard, as its’ predecessors, offers the hope that there is indeed a better life out there to those who wish to grab a hold of it. It provided to me a good feeling when DJ entered Heritage House and paid respect to all the Civil Rights leaders and influential people who paved the way for him to succeed. More movies need to focus on uplifting people rather than portraying their stereotypes and continuing the circle of despair. Yes, its a strained conceit, but Sylvain Whites movie does well to remind all of us of these powerful embodiments of resistance, motivation, and truth.

If you do need lighting, try to avoid directly illuminating the beach, using a shade to minimize the amount of light shining in the area. If you spot sea turtles in the water, stay at least 50 yards away. If they are close to your boat, put your engine on neutral or turn it off until the turtles swim away. Change your fishing location if you spot sea turtles nearby or they show interest in your bait. And remember to collect all of your fishing gear and supplies once youre done, especially fishing line, hooks, and nets. If you do want to watch one, attend a sea turtle watch hosted by an organization, which would allow you to observe the sea turtles without disturbing them. Do not catch a baby turtle in an aquarium or bucket of water. This will use up the energy they need to swim to the ocean after they emerge from their nest. Reduce Your Carbon Footprint Global warming can skew the gender ratios of sea turtles, as well as the distribution of predators and prey. Although climate change might seem like too big an issue to tackle, there are many steps you can personally take to reduce global warming. Adopt a Sea Turtle Support sea turtle conservation efforts by â€Å"adopting a sea turtle† or making a donation to a wildlife conservation program that monitors and helps satellite-tracked turtles. You can also â€Å"adopt a nest† during nesting season. Avoid Beach Activities at Night Try to avoid walking on the beach at night during the summer, as this may frighten nesting turtles back into the sea. To help make it easier for turtles to navigate the beach, you can also remove beach furniture and other equipment from the beach before the nighttime, as turtles may become caught in them or become disoriented. Help Spread Awareness There are many ways you can help make a positive change for sea turtles. One main way is through education. You can help educate your local neighborhood or school by giving presentations, and tell people about the cause during conversations. This follows that women are expected to maintain their physical beauty to remain acceptable in society. Women’s worth in society is often reflected in how ‘beautiful’ they are perceived by society, unlike men whose worth does not depend on their beauty (Shaw and Lee). This is often referred to as DOUBLE STANDARDS. This concept has been used by Victoria’s Secret which uses glamorous images of women throughout its store so that w omen feel they will look like those idealized images after wearing those products. A small fraction of women who are physically attractive are used throughout in-store photography which reflects the stereotypical belief that a woman’s worth is through her body and attractiveness. This is linked to the OBJECTIFICATION of women whereby their bodies are considered separate from the context (Shaw and Lee). This, in turn, is closely tied to the fragmentation of women when their bodies are separated from their personalities and are thought to represent the woman. The way in which specific body parts are highlighted in the photography reflects how the woman as a whole is discarded and how women are considered as objects that can be touched, â€Å"ogled† or even bought. The focus on specific parts such as breasts and other genital organs in these ads is reflective of the SURGERIES including VAGINAL SURGERIES that women undergo in the name of beauty. Craig Ventors First Cell

Cr Craig Ventor first first self-replicating, synthetic bacterial cell ROCKVILLE, MD and San Diego, CA (May 20, 2010)— Researchers at the J. Craig Venter Institute (JCVI), a not-for-profit genomic research organization, published results today describing the successful construction of the first self-replicating, synthetic bacterial cell. The team synthesized the 1. 08 million base pair chromosome of a modified Mycoplasma mycoides genome. The synthetic cell is called Mycoplasma mycoides JCVI-syn1. and is the proof of principle that genomes can be designed in the computer, chemically made in the laboratory and transplanted into a recipient cell to produce a new self-replicating cell controlled only by the synthetic genome. This research will be published by Daniel Gibson et al in the May 20th edition of Science Express and will appear in an upcoming print issue of Science. â€Å"For nearly 15 years Ham Smith, Clyde Hutchison and the rest of our team have been working toward thi s publication today–the successful completion of our work to construct a bacterial cell that is fully controlled by a synthetic genome,† said J.Craig Venter, Ph. D. , founder and president, JCVI and senior author on the paper. â€Å"We have been consumed by this research, but we have also been equally focused on addressing the societal implications of what we believe will be one of the most powerful technologies and industrial drivers for societal good. We look forward to continued review and dialogue about the important applications of this work to ensure that it is used for the benefit of all. † According to Dr.Smith, â€Å"With this first synthetic bacterial cell and the new tools and technologies we developed to successfully complete this project, we now have the means to dissect the genetic instruction set of a bacterial cell to see and understand how it really works. † To complete this final stage in the nearly 15 year process to construct and boot u p a synthetic cell, JCVI scientists began with the accurate, digitized genome of the bacterium, M. mycoides. The team designed 1,078 specific cassettes of DNA that were 1,080 base pairs long. These cassettes were designed so that the ends of each DNA cassette overlapped each of its neighbors by 80bp.The cassettes were made according to JCVI’s specifications by the DNA synthesis company, Blue Heron Biotechnology. The JCVI team employed a three stage process using their previously described yeast assembly system to build the genome using the 1,078 cassettes. The first stage involved taking 10 cassettes of DNA at a time to build 110, 10,000 bp segments. In the second stage, these 10,000 bp segments are taken 10 at a time to produce eleven, 100,000 bp segments. In the final step, all 11, 100 kb segments were assembled into the complete synthetic genome in yeast cells and grown as a yeast artificial chromosome.The complete synthetic M. mycoides genome was isolated from the yeast c ell and transplanted into Mycoplasma capricolum recipient cells that have had the genes for its restriction enzyme removed. The synthetic genome DNA was transcribed into messenger RNA, which in turn was translated into new proteins. The M. capricolum genome was either destroyed by M. mycoides restriction enzymes or was lost during cell replication. After two days viable M. mycoides cells, which contained only synthetic DNA, were clearly visible on petri dishes containing bacterial growth medium.The initial synthesis of the synthetic genome did not result in any viable cells so the JCVI team developed an error correction method to test that each cassette they constructed was biologically functional. They did this by using a combination of 100 kb natural and synthetic segments of DNA to produce semi-synthetic genomes. This approach allowed for the testing of each synthetic segment in combination with 10 natural segments for their capacity to be transplanted and form new cells. Ten out of 11 synthetic fragments resulted in viable cells; therefore the team narrowed the issue down to a single 100 kb cassette.DNA sequencing revealed that a single base pair deletion in an essential gene was responsible for the unsuccessful transplants. Once this one base pair error was corrected, the first viable synthetic cell was produced. Dr. Gibson stated, â€Å"To produce a synthetic cell, our group had to learn how to sequence, synthesize, and transplant genomes. Many hurdles had to be overcome, but we are now able to combine all of these steps to produce synthetic cells in the laboratory. † He added, â€Å"We can now begin working on our ultimate objective of synthesizing a minimal cell containing only the genes necessary to sustain life in its simplest form.This will help us better understand how cells work. † This publication represents the construction of the largest synthetic molecule of a defined structure; the genome is almost double the size of the previou s Mycoplasma genitalium synthesis. With this successful proof of principle, the group will now work on creating a minimal genome, which has been a goal since 1995. They will do this by whittling away at the synthetic genome and repeating transplantation experiments until no more genes can be disrupted and the genome is as small as possible. This minimal cell will be a platform for analyzing the function of every essential gene in a cell.According to Dr. Hutchison, â€Å"To me the most remarkable thing about our synthetic cell is that its genome was designed in the computer and brought to life through chemical synthesis, without using any pieces of natural DNA. This involved developing many new and useful methods along the way. We have assembled an amazing group of scientists that have made this possible. † As in the team’s 2008 publication in which they described the successful synthesis of the M. genitalium genome, they designed and inserted into the genome what they called watermarks.These are specifically designed segments of DNA that use the â€Å"alphabet† of genes and proteins that enable the researcher to spell out words and phrases. The watermarks are an essential means to prove that the genome is synthetic and not native, and to identify the laboratory of origin. Encoded in the watermarks is a new DNA code for writing words, sentences and numbers. In addition to the new code there is a web address to send emails to if you can successfully decode the new code, the names of 46 authors and other key contributors and three quotations: â€Å"TO LIVE, TO ERR, TO FALL, TO TRIUMPH, TO RECREATE LIFE OUT OFLIFE. † – JAMES JOYCE; â€Å"SEE THINGS NOT AS THEY ARE, BUT AS THEY MIGHT BE. †-A quote from the book, â€Å"American Prometheus†; â€Å"WHAT I CANNOT BUILD, I CANNOT UNDERSTAND. † – RICHARD FEYNMAN. The JCVI scientists envision that the knowledge gained by constructing this first self-replicatin g synthetic cell, coupled with decreasing costs for DNA synthesis, will give rise to wider use of this powerful technology. This will undoubtedly lead to the development of many important applications and products including biofuels, vaccines, pharmaceuticals, clean water and food products.The group continues to drive and support ethical discussion and review to ensure a positive outcome for society. Funding for this research came from Synthetic Genomics Inc. , a company co-founded by Drs. Venter and Smith. Background The research published today was made possible by previous breakthroughs at JCVI. In 2007 the team published results from the transplantation of the native M. mycoides genome into the M. capricolum cell which resulted in the M. capricolum cell being transformed into M. mycoides. This work established the notion that DNA is the software of life and that DNA dictates the cell phenotype.In 2008 the same team reported on the construction of the first synthetic bacterial ge nome by assembling DNA fragments made from the four chemicals of life—ACGT. The final assembly of DNA fragments into the whole genome was performed in yeast by making use of the yeast genetic systems. However, when the team attempted to transplant the synthetic bacterial genome out of yeast and into a recipient bacterial cell, viable transplants could not be recovered. Ethical Considerations: Since the beginning of the quest to understand and build a synthetic genome, Dr.Venter and his team have been concerned with the societal issues surrounding the work. In 1995 while the team was doing the research on the minimal genome, the work underwent significant ethical review by a panel of experts at the University of Pennsylvania (Cho et al, Science December 1999:Vol. 286. no. 5447, pp. 2087 – 2090). The bioethical group's independent deliberations, published at the same time as the scientific minimal genome research, resulted in a unanimous decision that there were no stron g ethical reasons why the work should not continue as long as the scientists involved continued to engage public discussion. Dr.Venter and the team at JCVI continue to work with bioethicists, outside policy groups, legislative members and staff, and the public to encourage discussion and understanding about the societal implications of their work and the field of synthetic genomics generally. As such, the JCVI’s policy team, along with the Center for Strategic & International Studies (CSIS), and the Massachusetts Institute of Technology (MIT), were funded by a grant from the Alfred P. Sloan Foundation for a 20-month study that explored the risks and benefits of this emerging technology, as well as possible safeguards to prevent abuse, including bioterrorism.After several workshops and public sessions the group published a report in October 2007 outlining options for the field and its researchers. Most recently in December of 2008, JCVI received funding from the Alfred P. Sloa n Foundation to examine ethical and societal concerns that are associated with the developing science of synthetic genomics. Air Travel.   Dr. Hans von Ohain and Sir Frank Whittle are both recognized as being the co-inventors of the jet engine. Each worked separately and knew nothing of the other’s work. Hans von Ohain is considered the designer of the first operational turbojet engine. Frank Whittle was the first to register a patent for the turbojet engine in 1930. Hans von Ohain was granted a patent for his turbojet engine in 1936. However, Hans von Ohain’s jet was the first to fly in 1939. Frank Whittle’s jet first flew in 1941.   The development of pressurised cabins meant that there was a supply of air which then they could fly higher and get out of the troposphere where all the weather conditions affected the flight. As without the pressurised cabin, flights couldn’t go above in to the stratosphere, into the thin air as there would be no air supply. This meant flights are more enjoyable and quicker, therefore more appealing to the public. Aircrafts can now reach heights of 30,000- 40,000 ft, whereas they could only reach heights of 15,000 ft. The Boeing 747, also popularly known as the jumbo jet, is the second largest passenger airliner after Airbus A380, but it remains the largest aircraft in commercial service. The 747 was born out of the explosion of the popularity of air travel in the 1960s. The enormous popularity of the Boeing 707 had revolutionised long distance travel in the world, and had began the concept of the global travel made possible by the jet revolution. The original design was a full length double decker. However, following issues with proposed evacuation routes, this idea was scrapped in favour of a wide-body design. Concorde 001 took off for the first test flight from Toulouse on March 2nd 1969 and the first supersonic flight followed on October 1st. As the flight program of the first development aircraft progressed as planned, 001 started off on a sales and demonstration tour beginning on September 4th 1971. Not until June 2nd 1972 did Concorde 002 follow suit with a sales tour of the Middle and Far East. Concorde 002 made the first visit to the United States in 1973, landing at the new Dallas/Fort Worth International Airport to commemorate its opening. Technological features. Sea Travel. * Sea travel has changed since the 2nd world war from the main form of global passenger transport to shorter-journey car ferries and cruises. This is because people use to go on a ferry to America because flying was very uncomfortable as technology was only developing. But when the jet engine and pressurised cabin were developed the need to go on the ferry was decreased as people could fly over instead, taking less time. This meant the ferry was not needed and so it became a cruise instead of like a `bus service`. * Ferries have become more popular due to the capacity of people they can get on them. This means that prices are reduced so it is more affordable for more people. Also as car ownership has increased the need for ferries which can hold cars is more important. Expedia is a virtual online travel agent which means you can book everything from you flight to hotel. Their aim is to replace high street travel agents.   Expedia also works with a GDS to get the results they want for their costumers. Holiday camps. In the 1950`s Butlins was very popular and was the main idea for a family holiday. This was the case then, when the concept of holidays was restricted to only the UK. Nine UK camps were built. They can be divided into three distinct eras: The pre-war camps of Skegness (1936) & Clacton (1938), the wartime camps of Filey, Pwllheli and Ayr and the post-war camps of Mosney (1948), Bognor (1960), Minehead (1962) and Barry (1966). There were already quite a few holiday camps in existence but Billy Butiln took the concept to a whole new level by making them far bigger and by offering a previously unheard of range of entertainment and activities. The main centre building offers everything from restaurants and bars to swimming pools all under one roof. Everything is very much more sophisticated than Butlins and is suited to families rather than single people. The family is a main target for travel and tourism and so centre parcs is making the most of the money that families have by accommodating for them.
